马鞭草C部位对人绒毛膜癌JAR细胞体外抑制作用的研究

    【摘要】 目的  探讨马鞭草提取液C部位对人绒毛膜癌JAR细胞体外抑制作用。 方法  MTT法测定不同浓度C部位在不同时段对JAR细胞的增殖抑制率;HCG单克隆酶免定量法检测马鞭草C部位对JAR细胞分泌HCG的影响;透射电镜观察细胞超微结构变化。 结果  JAR细胞经马鞭草C部位作用后,细胞增殖抑制率随着时间的延长和浓度的增加逐渐增大,72h的40g/L剂量组抑制最明显,抑制率为65%;HCG分泌受到明显抑制,呈剂量和作用时间依赖性;马鞭草C部位可引起细胞数目减少并萎缩;电镜下见到明显的凋亡小体。 结论  马鞭草提取液C部位对人绒毛膜癌JAR细胞增殖及分泌HCG有明显抑制作用,并可诱导JAR细胞凋亡。

    Inhibitory effects of the part C in alcohol extract of Verbena officinalis on

    human choriocarcinoma JAR cell line in vitro

    【Abstract】 Objective To investigate the effect of the part C in alcohol extract of Verbena officinalis on hu-man choriocarcinoma JAR cell line.Methods JAR cells were determined in the proliferation-inhibition ratio of the part C with vary chroma during different period of time using MTT method.The productions of HCG were detected during different time;morphological changes of JAR cells were observed and lastly the changes of ultrastructure were observed in different action time.Results The inhibition rate of experimental groups was more than that in control group and increased according to increasement of time and concentration.In72hours40g/L,there was a significant control and its inhibition was65%.Verbena officinals C could significantly inhibit the productions of HCG and the inhibitories effect were dose-and-time-dependent.The incubated tumor cells were decreased in number and be-came atrophic when they were treated with the part C.Apoptosis body were observed in experimental model of JAR.Conclusion The part C in alcohol extract of Verbena officinalis is a highly effective antineoplastic agent to choriocar-cinoma cells,manifestating time-concentration effect and could induce apoptosis.

    马鞭草(Verbena officinalis L.)系马鞭草科马鞭草属植物,分布较广。其始载于《名医别录》,具有活血散瘀、截疟、解毒、利水消肿等功效,在我国的汉族、彝族、傣族等25个民族广泛使用。主要被用于治疗支原体肺炎、急性乳腺炎等许多炎症性疾病,还作为防治肾癌、前列腺癌、晚期肝癌的首选药物。近年来,关于中草药诱导肿瘤细胞凋亡的研究日益增多,它们以与其他化疗药物不同的作用机制受到人们的重视。马鞭草作为一种普通的中草药,已被证实具有兴奋大鼠离体子宫肌条的作用和抗早孕作用 [1,2] 。本研究室也已证明马鞭草醇提液对人绒毛膜癌JAR细胞具有特异性的抑制作用 [3] 。徐昌芬 [4] 等从醇提液中筛选出一有效C部位,并提示马鞭草可能诱导JAR细胞凋亡。本文就马鞭草C部位在体外对人绒毛膜癌JAR细胞抑制作用初步研究探讨如下。

    1 材料与方法

    1.1 材料 绒毛膜癌JAR细胞株由上海细胞生物学研究所提供,DMEM培养基(美国GIBCOL公司产品),胎牛血清由杭州四季青生物工程公司供应,马鞭草C部位由江苏省中医药研究所提取分离,每毫升含生药1g,无菌抽滤后用含10%胎牛血清的DMEM培养液稀释到所需浓度;溴化钾噻唑基四唑(MTT)为Fluka公司产品;HCG单克隆酶免定量检测试剂盒由中国协和医科大学基础所提供,HCG标准品为Sigma公司产品;CO 2 培养箱:Precision scientific,USA;日本OLYMPUS倒置显微镜;奥地利SUN ZISE酶标仪;EM-1010为日本日立公司产品,LKB-V超薄切片机。
