A tractAIM: To study the effect of sodium butyrate in
combination withchuanhuning on HCT-8cell line proliferation.
METHODS:Inhibition of HCT-8 cell line by sodium butyric acid in
combination withchuanhuning was detected by MTT a ay and growth
curve, and apoptosis wasdetermined by morphological a ay and flow
cytometry (FCM). Apoptoticcells were o erved electro-
microscopically.
RESULTS:Sodium butyric acid showed inhibitory effect on the
proliferation of HCT-8cell line in dose-dependent and time-dependent
ma er. The inhibitoryrates were 15.7 %,20.3 %, and 33.3 % (P <0.01)
in differentgrou . Differentiation and apoptosis were o erved
under electronicmicroscope. Sub-G1 peak was detected by FCM. Cell
cycle was blocked in hase. The apoptotic rate of combinedgroup 1
were 23.5 %, 48.6 % at 24 h, and 48 h, and the apoptotic rate
ofcombined group 2 were 30.8 %, 54.2 % at 24 h, and 48 h (P <0.01).
CONCLUSION:Sodium butyric acid can induce apoptosis and
differentiation of HCT-8cells of human colorectal carcinoma, and
inhibit proliferation of HCT-8cells. Apoptotic rate was
significantly increased when sodium butyric acidwas combined with
Chuanhuning.
